Adenylate cyclase toxin (CyaA) of Bordetella pertussis belongs to the RTX family of toxins. These toxins are characterized by a series of glycine- and aspartate-rich nonapeptide repeats located at the C-terminal half of the toxin molecules. For activity, RTX toxins require Ca2+ , which is bound through the repeat region. Here, we identified a stretch of 15 amino acids (block A) that is located C-terminally to the repeat region and is essential for the toxic activity of CyaA. Block A is required for the insertion of CyaA into the plasma membranes of host cells. Mixing of a short polypeptide composed of block A and eight Ca2+ binding repeats with a mutant of CyaA lacking block A restores toxic activity fully. This in vitro interpolypeptide complementation is achieved only when block A is present together with the Ca2+ binding repeats on the same polypeptide. Neither a short polypeptide composed of block A only nor a polypeptide consisting of eight Ca2+ binding repeats, or a mixture of these two polypeptides, complement toxic activity. It is suggested that functional complementation occurs because of binding between the Ca2+ binding repeats of the short C-terminal polypeptide and the Ca2+ binding repeats of the CyaA mutant lacking block A. 相似文献

A large scale biomanipulation of pelagic fish by trawling was started in the eutrophic Enonselkä basin (26 km2) of Lake Vesijärvi to improve the water quality which had remained poor in spite of the termination of nutrient loading. The distribution and density of the fish were studied by hydroacoustics before and during the removal. The initial annual fish density varied between 13 000–21 000 fish ha–1 in the study area in August 1984–89. The mass-removal of the fish by pelagic trawling took place in 1989–1992. The catch varied annually between 64 and 92 kg ha–1. Roach (Rutilus rutilus (L.)) and smelt (Osmerus eperlanus (L.)) accounted for c. 85% of the weight of the catch. The mass-removal decreased the pelagic fish density in the Enonselkä basin during the trawling. An increase in the density was observed after the trawling ceased in these years, and the initial density level was reached within one month. The density level after mass-removal remained high compared with oligotrophic lakes. The pelagic fish had a diurnal ascending trend with the decreasing light intensity in August, and the fish were significantly (p < 0.01) higher in the water mass in temperature non-stratified water in autumn than in stratified water in summer. The fish were somewhat deeper in the years of mass-removal than before it. Mass-removal did not affect the individual length of the echosurveyed fish. The development of frost hardiness in forest trees is describedby a dynamic model in which the input variables are the prevailingenvironmental conditions and the developmental stage of trees.The assumption of the model is that for each temperature andphotoperiod there is a discrete stationary level of frost hardiness,which is attained if these environmental factors remain constant.The dependence of the stationary level on temperature and photoperiodis assumed to be piece-wise linear and additive. The rate ofacclimation, i.e. frost hardening or dehardening, is describedas a second-order dynamic process with two time constants, thesecond of which changes depending on the stage of the annualdevelopment of the trees. The frost hardiness model was calibratedand tested using experimental data from Douglas fir [Pseudotsugamenziesii var. glauca (Beissn.) Franco] seedlings. The potential contribution of fucosyltransferases to the overexpression of sialyl-Lex antigen was investigated in the colon carcinoma cell line HT-29 and in human colon carcinoma tissue. In HT-29 cells as well as in normal or malignant colonic tissues Fuc-TIII, Fuc-TIV, Fuc-TVI but not Fuc-TV nor Fuc-TVII were detectable after RT-PCR. Sodium butyrate treatment of HT-29 cells increased (to about 200%) and DMSO treatment decreased (to about 20%) the expression of sialyl-Lex. This modulation of sialyl-Lex was concomitant with the analogous increase/decrease of mRNA of Fuc-TIII but not Fuc-TIV. Fuc-TVI was not detectable by Northern blotting in HT-29 cells. In six human colon carcinomas which exhibited strong overexpression of sialyl-Lex, the expression of Fuc-TIII-mRNA was the same or lower than in the corresponding normal colonic tissue. Thus Fuc-TIII expression may be affecting the expression of the sialyl-Lex moiety in HT-29 cells but not in human colon carcinoma tissue. 相似文献

The typification of names in the genus Cyperus was done as part of an ongoing study of the section Arenarii. The latest monograph on the genus by Kükenthal (1936), accepted eight species in the section Bobartia (= Arenarii ), and a number of lower level taxa, which are treated here either as distinct species or synonyms. The taxonomic treatment of the core species, C. conglomerates Rottb., turned out to be especially confusing, which is reflected in the past identifications of the material, and consequently, has created wrong interpretations on the distributions. C. jeminicus Rottb. has similarly caused a lot of confusion. Currently we recognize 26 species in the section, with two subspecies in C. conglomerate. Most names in the section have not earlier been typified. Here we designate lectotypes for 33 names, three epitypes, and one neotype. Background
Wnt11 is a member of the Wnt family of secreted signals controlling the early steps in ureteric bud (UB) branching. Due to the reported lethality of Wnt11 knockout embryos in utero, its role in later mammalian kidney organogenesis remains open. The presence of Wnt11 in the emerging tubular system suggests that it may have certain roles later in the development of the epithelial ductal system.Results
The Wnt11 knockout allele was backcrossed with the C57Bl6 strain for several generations to address possible differences in penetrance of the kidney phenotypes. Strikingly, around one third of the null mice with this inbred background survived to the postnatal stages. Many of them also reached adulthood, but urine and plasma analyses pointed out to compromised kidney function. Consistent with these data the tubules of the C57Bl6 Wnt11 ?/? mice appeared to be enlarged, and the optical projection tomography indicated changes in tubular convolution. Moreover, the C57Bl6 Wnt11 ?/? mice developed secondary glomerular cysts not observed in the controls. The failure of Wnt11 signaling reduced the expression of several genes implicated in kidney development, such as Wnt9b, Six2, Foxd1 and Hox10. Also Dvl2, an important PCP pathway component, was downregulated by more than 90 % due to Wnt11 deficiency in both the E16.5 and NB kidneys. Since all these genes take part in the control of UB, nephron and stromal progenitor cell differentiation, their disrupted expression may contribute to the observed anomalies in the kidney tubular system caused by Wnt11 deficiency.Conclusions
The Wnt11 signal has roles at the later stages of kidney development, namely in coordinating the development of the tubular system. The C57Bl6 Wnt11 ?/? mouse generated here provides a model for studying the mechanisms behind tubular anomalies and glomerular cyst formation.89.

The dextransucrase gene from Weissella confusa Cab3, having an open reading frame of 4.2?kb coding for 1,402?amino acids, was amplified, cloned, and expressed in Lactococcus lactis. The recombinant dextransucrase, WcCab3-rDSR was expressed as extracellular enzyme in M17 medium with a specific activity of 1.5?U/mg which after purification by PEG-400 fractionation gave 6.1?U/mg resulting in 4-fold purification. WcCab3-rDSR was expressed as soluble and homogeneous protein of molecular mass, approximately, 180?kDa as analyzed by SDS-PAGE. It displayed maximum enzyme activity at 35°C at pH 5.0 in 50?mM sodium acetate buffer. WcCab3-rDSR gave Km of 6.2?mM and Vm of 6.3?µmol/min/mg. The characterization of dextran synthesized by WcCab3-rDSR by Fourier transform infrared and nuclear magnetic resonance spectroscopic analyses revealed the structural similarities with the dextran produced by the native dextransucrase. The modeled structure of WcCab3-rDSR using the crystal structures of dextransucrase from Lactobacillus reuteri (protein data bank, PDB id: 3HZ3) and Streptococcus mutans (PDB id: 3AIB) as templates depicted the presence of different domains such as A, B, C, IV, and V. The domains A and B are circularly permuted in nature having (β/α)8 triose phosphate isomerase-barrel fold making the catalytic core of WcCab3-rDSR. The structure superposition and multiple sequence alignment analyses of WcCab3-rDSR with available structures of enzymes from family 70 GH suggested that the amino acid residue Asp510 acts as a nucleophile, Glu548 acts as a catalytic acid/base, whereas Asp621 acts as a transition-state stabilizer and these residues are found to be conserved within the family. 相似文献

Summary The gene coding forBordetella pertussis P.69 protein was cloned and expressed inBacillus subtilis. The expression vector contained the promoter region and the sequence coding for the whole or truncated signal sequence of the -amylase gene fromB. amyloliquefaciens. Using either construction the level of expression was relatively low and the protein was found in the particulate fraction.
